We have been using the TissueGnostics microscope platform and hardware, and the TissueFax and HistoQuest and TissueQuest software applications for 3 years now. The data we have generated has allowed us to gain insights into the DNA-damage repair mechanisms, examine the modes of cell death and to quantify the cell cycle effects that underpin the efficacy of our antibody targeted therapies. I have worked in the field of academic and routine pathology and image analysis for almost forty years and there is no doubt in my mind that the TissueGnostics system provides the best option for critical examination of dye-stained sections, immunohistochemistry and immunofluorescence at all levels. It has the ability to acquire images from multiple histological sections rapidly, efficiently and reliably, to define, segment and threshold regions of interest for analysis, it can measure events objectively and reproducibly across multiple parameters, and the data can be displayed in an innovative and easy to interrogate manner. It has the unique ability to track single data point events from the display scatter-grams, back to the image to enquire about the position and type of cell within the tissue from which that measurement was generated. The level of interaction with the software programmes requires training and practice, and some learning of the fundamentals of image analysis is necessary, but users do not need previous programming experience to work easily with the system to gain optimal outputs – they just need to allocate time and practice. Our latest submitted manuscript to ‘Cancer Research’ contains some important data from quantitation of immunohistochemistry using our TissueGnostics system, and has allowed us to confidently propose a mechanism of action for our combined therapy, in a way that would not have been possible from our previous image analysis platforms. The company’s team of support engineers and software programmers are always available to advise on any maintenance issues and software fixes, and they will always provide confirmation that the techniques being used by the investigator/researcher are appropriate and optimal. As part of the Center for iPS Cell Research and Application (CiRA) of Kyoto University, our research group aims to develop a cell replacement therapy for Parkinson’s disease using iPS cells. Our main work includes inducing and isolating transplantable neurons from iPS cells, and evaluating the grafts after transplantation of these cells into animal brains. Since the TissueFAXS system has been introduced into the center a few years ago, we have benefitted greatly from it.

For example, large areas of tissue can be easily sampled with good speed and quality. We are particularly impressed with the beautiful overview images of brain sections obtained for multiple fluorescent channels and also for brightfield. What can be done next with the obtained images using TissueQuest and HistoQuest analysis software was even more incredible.

The system provides quantitative data that are objective and reliable for immunofluorescence studies, and saved us a great deal of time and effort for manually counting cells on many sample slides. In order to count cells in the most suitable way for each sample, it first makes sure that individual cells are segmented accurately. They are then represented as dots on scattergrams according to the intensity of fluorescence, just like for FACS dot plots, and lets us define positive cells by showing the location of corresponding dot on scattergram for any selected cell on the fluorescent image. Being used effectively for appropriate samples, it is undeniable that the TissueFAXS system has greatly improved the efficiency of our imaging work so that we can deal with hundreds of samples comfortably and confidently in our challenging project.
